Main difference is that in Otoko no Ko, the female lead is male - Both have assertive, flirtatious male leads - Focused on Slice of Life aspect

Both have the same heroine a Boy, friends since childhood. in terms of story and Romance development Koisuru (Otome) no Tsukurikata is way better, and you'll forget that Hiura is a boy when you read it. the storyline is also relaxed focused on Slice of Life aspect and the number of panels for each chapter is not too much so you don't get bored while reading it.

Cute slice of life takes on a couple where both are men but one cross-dresses as a hobby. Nobody is mocked, everybody is accepted and the stories are pretty wholesome overall. Both also have a super-moe and simplistic artstyle.

You can treat Otoko no Ko as an unofficial sequel to Boy skirt.

Both mangas focus on homosexual relationships with crossdressing. And both don't touch on very serious topics (especially Prunus Girl), which makes them very easy to read.

They both involve a couple, both who are guys, with one dressing up as a girl. While the relationships of both stories are cute and wholesome, they also delve into gender and sexuality, more so in Fukakai than in Otoko. The crossdresser in Otoko considers himself as a guy while the one in Fukakai considered themselves as non-binary, so you can see that it delves into those topics.
