In a more metaphorical sense, running indicates that we will be having an adventure throughout the series.

In the technical animation aspect, it could just be that a running sequence is far easier to animate since they just have to repeat the same motion for most times.

There might also just be less to show from a romance anime, just the characters. You have to be realy creative to make interesting romance anime openings like Kaguya-sama. So just throw characters running to make people think stuff happens.
