For me, there were three,

Number 3: Kingseeker Frampt

After ringing the first bell, the one on top of the Gargoyle-Tower, you can hear a strange scraping sound at where, later Frampt emerges at the Firelink Shrine.
A sound, that wasn't there before...
And there's even something written in the subtitle...
this totally freaked me out.
I got totally nervous and searched the place for nearly a half hour to ensure that there's no boss, that whould jump my back, as soon as i turend around.
The last time a game made me that nervous, was The Ledgend of Zelda: Ocarina of Time, back in 1998... when i was 11...

Number 2: The Gaping Dragons emerge.

At first i thought, "Pfft.... that's considered to be the next boss...?"
Then the dragon stood up and i saw more, than it's nose, which i had mistaken for it's head and thought "Dammit... why can't things ever be easy....?"
While the Gaping dragon might not be the most beautiful dragon, ever created in a video game, at least it's a very unique version of a dragon.
At least i have not seen many similar taken on a dragons appearence.
So, thumbs up up the creativity, that went into one of the most awesome ways of saying: "Women are evil!"

Number 1: My first arival in Anor Londo.

I went to the Bonfire and when i came back up to the balcony, and thought,
"Well... that's a really goreous view."
I stood there and did nothing but really enjoy the view for at least five minutes.
Seriously, it's been years, since a game managed to stun me by a mere background.

I think the last game was actually Ocarina of Time, too...
speaking of which... for me Dark Souls has a very similar feeling to Ocarina of Time...

So, my
Number 0(carina of time)

The whole game made me feel like i am play Ocarina of Time again, in a new and exciting way.
I'm not sure if Dark Souls is able to claim the title of my all time favourite video game, only time will show...

#3: Tomb of Giants

Tomb of Giants....who can forget the brutal surprise buttsex they suffered while trying to navigate through a black hole filled with giant skeletons waiting to rape u. :l

#2: Ornstein and Smough

you walk into the fog and see a huge fat dude with an equally huge hammer waiting for you, looks like he'd be a handful on his own, then a badass lookin knight jumps down and joins him. At that point you think "aww f***" :l
imo the soundtrack that plays during this fight is f***ing epic and makes the fight all the more enjoyabe.
then when you beat one of em, you feel relaxed because your half way done and it should be easier with only one boss to fight....but no, the surviving boss promptly eats the other one's soul and goes all super saiyan on yur ass.

#1: Gwynevere

Who can forget her GIGANTIC RACK? :3

im sure im not the first, and i wont be the last, but when i first gazed upon her holy gazongas....i went to the bonfire and whipped out my binoculars and cast light spell for a better view :P

p.s.
loved the music for the final fight against gwyn....sad and gloomy, but appropriate and awesome! :3

4# It has to be killing Gwyn lord Of Cinder, killing the old dead husk of a god made me feel bad. Dark souls is the only game so far to make me feel bad for killing a boss.

3# Used the spell that disguises you as an object. They searched for ages, in the end they found me but it was pretty funny.

2# 3v1 Forest Hunter, i only used force push to defeat them all.

1# As a forest Hunter i was being ganged up by 2v1. I was holding against the host an luckily a red Phantom came to help me in time. We teamed up so it was a 2v2 and the match lasted for 5 minutes with our duo being victorious. The fight was so epic.

Killing both Gwyn and Priscilla. Not many games make you even care about the bosses you fight, but both of them give you a sense of '...what have I done?' Oh god why.' Especially with Priscilla being non-aggressive and all.

Going into the final boss thinking 'Oh god what horrible thing am I going to fight here?' and then seeing an old man, hollowed out and dying, fighting to his end, accompanied by a beautiful piano piece. Definiately not expected, but very much worth it.

Props to both Gaping Dragon and Ornstein & Smough for being pretty damn epic boss fights too. I also can't get Gaping Dragon's theme out of my head with it's weird instruments.
