Since the setting of the show/manga is in another world, are the characters
European/American or Japanese?

The setting is another world, so it's neither. I would say the clothing and architecture would not be out of place in some western aristocracy setting.

Technically if an anime series doesn't name a real world country or geographic location for its setting, then I take it as them not officially designating the characters any specific nationality, race, or ethnicity.

But in some cases it's heavily alluded to even if the names are changed and no real world place/people are labeled explicitly. Some are made with very strong historical allusions and parallels in mind. It's sometimes coded through the clothing, the cuisine, the nature/landscape, mannerisms/customs, or the look of the people.

I would say Shadows House is made to evoke a neo-Gothic sense. Like a Gothic mansion on a vast land holding by isolated European aristocrats in the countryside. For some reason what little they've showed of the landscape reminds me of Switzerland or a mountainous region of France, but I don't think the actual specific individual nationality is either all that important or even feasibly discernible based on the information given.

I believe general landed class European aristocracy maybe sometime in the Victorian period in a fairy tale-inspired world and aesthetic (seems to have some magical realism though as from what's been pictured of the outside world beyond the Shadows' family estate seems to me the rest of the world could be mostly or completely devoid of magic and supernatural entities and they're just segregated here without openly interacting with the human world, similar to the Harry Potter/Hogwarts situation).

the gothic lolita-esque clothing and fashion style + the architecture and overall setting seemed pretty reminiscent of the victorian era or rococo period. just from taking a quick look at the show you can definitely tell it's formulated on and inspired by europe, most likely either france or england, though the character's names lead me to believe the latter is more plausible.
