They are both similar in the fact that the female protagonists don't initially intend to fall for the main male. I felt that Lock On had a more fighting type feel while You're My Girlfriend was more slice-of-life, in a manner of speaking.
If you've read You're My Girlfriend, you'll like Lock On. (Or, of course, vice versa)
