The characters are exactly the same; one bookworm/otaku girl, one energetic dopey girl and one headstrong, glutonous girl; all very different young schoolgirls. Considering both are comedy seriese's, character type is the most importiant element, as it is sutiable the character who is thrown into comical scenario; both are series's were the comic element can take you anywhere, depending on the writers taste.

Similar comedy, slightly younger characters (or so I believe) and a little more ecchi. I found that Nichijou filled the void after Mitsudomoe S1 and S2 were finished airing.
Nichijou is by far superior in animation quality and humour although Mitsudomoe has the same feel and pacing, Mitsudomoe is a bit more on the odd sexual side of humour compared to Nichijou but both have really great comedic moments. If you are after something to sit bac, zone out and laugh too they are two peas in a pod and if you like one you should like the other.
