Both are comedy, slice-of-life anime, with a focus on a family of 3 sisters. Each individual has a stereotypic personality, which you can pretty much predict their responses/interactions to different situations. Both series will make you laugh out loud.
Comedy, slice of life, scenario based and can be adventerous are a trate both anime's share, aswell as the character types; the fact that both revolve around three very different sisters for a start:
- The oldest is a ring leader; in Minami-Ke, the oldest is said to be an ex gang memeber, whilst in Mitsudome, the oldest 'leads her class' and can be quite sinister
- The middle sister in both series's is hyperactive
- The youngest is quite intellectual, yet socially awkward, wears a aloff expression and is fairly quiet

Both have similar styles of humor. Mitsudomoe goes more along the ecchi line of things whereas Minami-ke follows a more straight up Slice of Life humor.
Linked by a name and a common environment, separated by the rest. Masahiko Ohta directs these two animations that portrays the daily lives of three so different sisters. "Minami-ke" is well-behaved, familiar, slow, a true "slice-of-life" that if it doesn't generate many laughs, at least can give the viewer a good amount of light and warm stories. In turn, the restless "Mitsudomoe" is often inconvenient and controversial with his heavy and questionable jokes of sexual nature, made around sixth grade students - but it is hugely funny at the same time that is so wrong,

Mitsudomoe and Minami-ke centers around the everyday life of three sisters, all of them with totally different personalities. Mitsudomoe is MUCH more twisted and dirtier than Minami-ke. Minami-ke has much more depth both in plot and carachters.
Both animes are slice-of-life comedies about three very different sisters.

talks about everyday life and hilarious jokes..
A sister trio that only knows of making life fun as a trio. There's no boring day for the Minami or Marui sisters. Everything is filled with some kind of wild event, trying to make the most out of life.
It's all the same business in both anime. School is funny. People coming over is funny. Interacting is funny. Mitsudomoe however is more surreal and crazy with what it pulls compared to Minami-ke's relatively tame and real life of sisters living with one another.
Take what you will, they are still both goof school comedies, quite episodic too.
