Everything else in this universe follows the usual rules of nasuverse and most of characters, theories and things work the exact same way as in other fate shows. Its equally canon.

Because its a multiverse. The original FSN visual novel itself had three distinctively different routes that contradicted each other, yet were equally canon. Nasuverse is more than one world and one setting - its a multiverse of different realities, akin to CLAMPverse, with those who know and have means to do so, traversing through those dimensions from one to other. That's why you can have stuff like cyberpunk Fate/Extra CCC or mahou shoujo like Prisma Illya.
Prisma Illya is alternate outcome of fate stuff and figuring out where and why it diverged is one of more enjoyable aspects of it. That and seeing very familiar characters make an appearance and be their usual badass selves.
So what we know of characters from previous series does not matter?

This show relies on that knowledge and touches quite heavily upon the whole multiverse theory of nasuverse. Knowing those characters and what happens to them in previous versions makes the storyline more complex. Without providing spoilers, all I can say is that the whole multiverse theory has quite a big importance to Prisma Illya storyline.
But, but, but its cutesy and non-serious
Both Madoka, gurren lagann and Nanoha shows started that way. If anything, Prisma Illya IS gurren lagann of mahou shoujo genre. Prisma Illya is not a traditional mahou shoujo genre show- its quite self-aware and quite a few things in it can get VERY brutal - I would like to remind you that FSN novel itself, too, starts off relatively lighter and gets darker with every route. It has enough grimdark elements in it, but it, as well as some characters KNOW the genre they are in.
In fact, I would say that there's a point when you would realize this is NOT a mahou shoujo genre story at all.

Nasuverse refers to a multiverse setting shared between various type-moon works. Its a vast multiverse of alternate realities in which type-moon works take place and it is connected by similar laws, rules, concepts and characters, all leading different lives in each timeline.
The characters of nasuverse are aware that it is a multiverse and some of them are capable of traveling between the worlds - the most common method being the "Second True Magic", Kaleidoscope, created by Grand Magister Zelretch.
At the same time, some of the concepts, like Heroic Spirits, are shared through the entire multiverse, due to the source of them being outside of each dimension.
Major parts of it are Tsukihime works, Fate franchise, Kara no Kyoukai and Mahou Tsukai No Yoru, but there's more to the nasuverse than just those though.

Tohsaka most likely IS dead, since Rin is living alone and still formed her obsessive-competitive personality that resulted from living alone.
Some of Zero masters will be revisited, some sooner, others latter. :)

But if the Kaleid timeline diverged from Fate0 before the 4th Holy Grail War starts, then the fate of Tohsaka and Kayneth cannot be so sure since they died in the war, esspecially for Tohsaka since he would be killed by Kirei who is not in Fuyuki city and who may not be Gilgamesh-fied.

Rin's personality is the indication of it. Tohsaka still died, one way or the other. He still met his end, resulting in Rin being obsessed with magus perfection and overly-judging towards others, just like in FSN
Whatever happened to Kariya and Tohsaka and Rin's Mother, it resulted in Rin's personality currently as well as
Rin having no idea who Sakura is
Kayneth HAS to be dead otherwise Waver would not hold that title.

we know that Iris and Kiri DID something and that Iris is the "guarantee" that HGW would be impossible and that their current "travels" are possibly connected to it. And that there are worlds where there's no grail war too(Ainsworth world had them experimenting with various replacements for grail)
Current events are 100% connected to it, though, considering when the card mess started

might be possible explanation, which could connect to current Drei plot, although I do not see how it would work exactly.
